In the development of dermal drug formulations and cosmetics, understanding the penetration properties of the active ingredients is crucial. Given that widespread methods, including tape stripping, lack in spatial resolution, while being time- and labour-intensive, Confocal Raman Microscopy is a promising alternative. In optimizing topically applied formulations, or the development of generic formulations, comparative in-situ measurements have a huge potential of saving time and resources. In this work, we show our approach to in-situ skin penetration analysis by confocal Raman Microscopy. To analyse feasibility of the approach, we used caffeine solutions as model vehicles and tested the effectiveness of 1,2-pentanediol as a penetration enhancer for delivery to the skin.Approximately 96% of patients with glioblastomas (GBM) have IDH1 wildtype GBMs, characterized by extremely poor prognosis, partly due to resistance to standard temozolomide treatment. O6-Methylguanine-DNA methyltransferase (MGMT) promoter methylation status is a crucial prognostic biomarker for alkylating chemotherapy resistance in patients with GBM. However, MGMT methylation status identification methods, where the tumor tissue is often undersampled, are time consuming and expensive. Currently, presurgical noninvasive imaging methods are used to identify biomarkers to predict MGMT methylation status. We evaluated a novel radiomics-based eXtreme Gradient Boosting (XGBoost) model to identify MGMT promoter methylation status in patients with IDH1 wildtype GBM. This retrospective study enrolled 53 patients with pathologically proven GBM and tested MGMT methylation and IDH1 status. Radiomics features were extracted from multimodality MRI and tested by F-score analysis to identify important features to improve our model. We identified nine radiomics features that reached an area under the curve of 0.896, which outperformed other classifiers reported previously. These features could be important biomarkers for identifying MGMT methylation status in IDH1 wildtype GBM. The combination of radiomics feature extraction and F-core feature selection significantly improved the performance of the XGBoost model, which may have implications for patient stratification and therapeutic strategy in GBM.Sonoran felids are threatened by drought and habitat fragmentation. Vector range expansion and anthropogenic factors such as habitat encroachment and climate change are altering viral evolutionary dynamics and exposure. However, little is known about the diversity of viruses present in these populations. Small felid populations with lower genetic diversity are likely to be most threatened with extinction by emerging diseases, as with other selective pressures, due to having less adaptive potential. We used a metagenomic approach to identify novel circoviruses, which may have a negative impact on the population viability, from confirmed bobcat (Lynx rufus) and puma (Puma concolor) scats collected in Sonora, Mexico. Given some circoviruses are known to cause disease in their hosts, such as porcine and avian circoviruses, we took a non-invasive approach using scat to identify circoviruses in free-roaming bobcats and puma. Three circovirus genomes were determined, and, based on the current species demarcation, they represent two novel species. Phylogenetic analyses reveal that one circovirus species is more closely related to rodent associated circoviruses and the other to bat associated circoviruses, sharing highest genome-wide pairwise identity of approximately 70% and 63%, respectively. At this time, it is unknown whether these scat-derived circoviruses infect felids, their prey, or another organism that might have had contact with the scat in the environment. Our end-to-end convolutional neural network consists of two stages, where at first a view-invariant trajectory descriptor for each body joint is generated from RGB images, and then the collection of trajectories for all joints are processed by an adapted, pre-trained 2D convolutional neural network (CNN) (e.g., VGG-19 or ResNeXt-50) to learn the relationship amongst the different body parts and deliver a score for the movement quality. We release the only publicly-available, multi-view, non-skeleton, non-mocap, rehabilitation movement dataset (QMAR), and provide results for both cross-subject and cross-view scenarios on this dataset.
