Methanol extract of the flowering aerial parts of Hypericum sinaicum Boiss. (ME) growing in Saint Catherine Protectorate (SKP), Egypt was analysed for its phenolic compounds profiling using HPLC and colorimetric methods. The total phenol content of ME was 158.60 ± 0.74 (μg GAE/mg D.E.), while the total flavonoid content was 70.91 ± 0.01 (μg QE/mg D.E.). HPLC analysis revealed that the highest flavonoid was naringenin (50.31 mg/g), while the highest phenolic acid was syringic acid (0.37 mg/g). The scavenging activity of ME was evaluated using DPPH assay with SC50 22.9 μg/ml and ABTS with SC50 13.10 μg/ml. ME produced dose - dependent and significant inhibition of edema at 4 hour of dose 200 mg/kg (78.55%) and 100 mg/kg (72.89%) to that of standard drug Indomethacin (86.94%). The current study interprets the anti-inflammatory and antioxidant potency of H. sinaicum.Aim Alterations in the microenvironment change the phenotypes of dental pulp stem cells (DPSCs). The role of complement component C5a in the differentiation of DPSCs is unknown, especially under oxygen-deprived conditions. The aim of this study was to determine the effect of C5a on the odontogenic differentiation of DPSCs under normoxia and hypoxia.Material and Methods Human DPSCs were subjected to odontogenic differentiation in osteogenic media and treated with the C5a receptor antagonist-W54011 under normal and hypoxic conditions (2% oxygen). Immunochemistry, western blot, and PCR analysis for the various odontogenic differentiation genes/proteins were performed.Results Our results demonstrated that C5a plays a positive role in the odontogenic differentiation of DPSCs. C5a receptor inhibition resulted in a significant decrease in odontogenic differentiation genes, such as DMP1, ON, RUNX2, DSPP compared with the control. This observation was further supported by the Western blot data for DSPP and DMP1 and immunohistochemical analysis. The hypoxic condition reversed this effect.Conclusions Our results demonstrate that C5a regulates the odontogenic DPSC differentiation under normoxia. Under hypoxia, C5a exerts a reversed function for DPSC differentiation. Taken together, we identified that C5a and oxygen levels are key initial signals during pulp inflammation to control the odontogenic differentiation of DPSCs, thereby, providing a mechanism for potential therapeutic interventions for dentin repair and vital tooth preservation. Selenium is an essential trace element for metabolism and is also a water pollutant. Selenium is adsorbed in the water by MNZ in this study first. Then the Brassica chinensis L. was planted in the soil which contains the MNZ loaded with selenium (MNZ-Se) to explore selenium uptake. MNZ-Se release tests in water and soil were also considered. The results showed the following (1) The maximum adsorption capacity of MNZ for selenium is 46.90 mg/g. (2) Water release experiments of MNZ-Se showed that regardless of how the pH of the aqueous solution changes, the trend of the release of selenium from MNZ-Se in aqueous solution is not affected and first decreases before stabilizing. (3) Soil release experiments of MNZ-Se showed that the selenium content in the soil increased and reached the concentration in the standard of selenium-rich soil. Addition amount and soil pH value will affect the release ratio. The release ratio of MNZ-Se in the water was higher than that in the soil. (4) With an increase in the soil MNZ-Se content, the selenium content in the soil and B. c increases. Above all, MZN can be a good medium for water pollutant removal and soil improvement.
