Using the broad-spectrum P2 antagonist suramin, the specific purinergic receptor P2Y2 (P2RY2) antagonist AR-C118925XX, and P2RY2-specific siRNA, we discovered that the HDM extract-induced IL-33 expression was mainly dependent on extracellular ATP/P2Y2 signaling mediated by transactivation of epidermal growth factor receptor, followed by activation of the ERK kinase signaling pathway. Moreover, HDM extract-induced release of 25-kDa IL-33 from the keratinocytes depended on an extracellular ATP/P2 signaling-mediated intracellular Ca2+ increase. Our study demonstrates the new mechanism controlling the induction and maturation of keratinocyte-produced IL-33 by HDM allergens, an innate immune process that might play a role in AD development or severity. When considering dermal administration of cosmeceuticals and/or drugs, the stratum corneum layer of the skin, has a barrier function that limits the penetration of active substances to the targeted skin tissues. Solid lipid nanoparticles/SLNs are colloidal carrier systems, which show superiority in dermal administration of cosmeceuticals/drugs. OBJECTIVE Understanding craniofacial growth and development is important for accurate diagnosis, treatment planning and post-treatment evaluation of orthodontic cases. Paramount to this is knowledge of the cranial base growth and development, since it is the foundation upon which the remaining facial structures develop. The purpose of this study was to analyse different anatomical structures in the posterior cranial base and determine its changes during the adolescent years (13-15 year of age) using CBCT. MATERIALS AND METHODS Sixty CBCT images of the adolescent population that received orthodontic treatment at two time-points (mean age T1 13.1 years/T2 14.6years) were used to assess growth related dimensional changes using 33 selected landmarks. Inter-rater, intra-rater and accuracy of 33 selected landmarks in the posterior cranial base and surrounding area were then evaluated via three-dimensional (3D) cone-beam computed tomography (CBCT). Linear differences were then calculated among the different landmarks to determine the changes present in the sample. RESULTS The selected landmarks in the posterior cranial base and surrounding area were found to be reliably and accurately located in 3D. Over the growth period studied (17.5months), minor statistically significant changes occurred, but they were deemed clinically irrelevant. CONCLUSIONS The studied landmarks in the posterior cranial base and surrounding area showed minor clinically insignificant changes over the study period. The observed changes could be attributed to measurement error. The posterior cranial base is deemed to be stable in all three dimensions of study during the adolescent growth period studied. BACKGROUND Connective tissue diseases (CTD) are frequently associated with interstitial lung disease (ILD), significantly impacting their morbidity and mortality. AIM Analyze the experience of an autoimmune specialized unit on treating CTD-ILD and characterize the population based on most frequent diseases, imaging patterns, lung function tests results, serology and treatment. Assess mortality and mortality predictors in these patients. METHODS Retrospective, descriptive and statistical analysis of the CTD-ILD patients followed up at an autoimmune diseases unit during a 6-year period. RESULTS Over the study period, 75 patients with CTD-ILD were treated with a mean follow-up of 49 ± 31 months. The most frequent CTD were systemic sclerosis and rheumatoid arthritis. ILD was diagnosed prior to CTD in 8% of patients and concomitantly in 35%. Nonspecific interstitial pneumonia was the CT pattern in 60% and 35% had an isolated diminished DLCO on lung function tests. Pulmonary hypertension was present in 12% and it was the single most important mortality predictor (OR 14.41, p = 0.006). Corticosteroids are the mainstay of treatment but biologics were prescribed in 39% of the patients (mostly tocilizumab and rituximab). Two scleroderma patients were recently treated with nintedanib. CONCLUSIONS ILD is a potential complication of every CTD and can impose a dramatic burden on these patients. The clinical relevance of ILD together with their early expression in the course of the disease underlines the importance of the presence of chest physicians in these units. OBJECTIVE To examine any correlations between tumor maximum standard uptake values (SUVmax) in positron emission tomography-computed tomography (PET-CT) and homogeneous/heterogeneous tumor FDG uptake in PET-CT, and the diagnostic success of the procedure in thoracic ultrasonography (US)-guided transthoracic fine needle aspiration biopsy (TFNAB). METHODS The files of patients who underwent thoracic US-guided TFNAB between 2013 and 2018 were examined. Patients who underwent thoracic US-guided TFNAB and were diagnosed as having primary lung cancer were considered as the US-TFNAB diagnostic group. Patients whose disease was diagnosed as primary lung cancer using a different diagnostic method (e.g. CT-guided biopsies, fiberoptic bronchoscopy) due to a lack of diagnosis despite undergoing thoracic US-guided TFNAB were allocated to the US-TFNAB non-diagnostic group. The clinical and radiologic characteristics and PET-CT parameters of the two groups were compared. RESULTS A total of 104 patients were included in the study; 79 (76%) patients whose disease was diagnosed using US-guided TFNAB, and 25 (24%) patients whose primary lung cancer could not be diagnosed with US-guided TFNAB.
