Hepatocellular carcinoma (HCC) is a liver tumor that usually arises in patients with cirrhosis. Hepatic stellate cells are key players in the progression of HCC, as they create a fibrotic micro-environment and produce growth factors and cytokines that enhance tumor cell proliferation and migration. We assessed the role of endoplasmic reticulum (ER) stress in the cross-talk between stellate cells and HCC cells. Mice with a fibrotic HCC were treated with the IRE1α-inhibitor 4μ8C, which reduced tumor burden and collagen deposition. By co-culturing HCC-cells with stellate cells, we found that HCC-cells activate IREα in stellate cells, thereby contributing to their activation. Inhibiting IRE1α blocked stellate cell activation, which then decreased proliferation and migration of tumor cells in different in vitro 2D and 3D co-cultures. In addition, we also observed cell-line-specific direct effects of inhibiting IRE1α in tumor cells.In fission yeast, the septation initiation network (SIN) ensures temporal coordination between actomyosin ring (CAR) constriction with membrane ingression and septum synthesis. However, questions remain about CAR regulation under stress conditions. We show that Rgf1p (Rho1p GEF), participates in a delay of cytokinesis under cell wall stress (blankophor, BP). BP did not interfere with CAR assembly or the rate of CAR constriction, but did delay the onset of constriction in the wild type cells but not in the rgf1Δ cells. This delay was also abolished in the absence of Pmk1p, the MAPK of the cell integrity pathway (CIP), leading to premature abscission and a multi-septated phenotype. Moreover, cytokinesis delay correlates with maintained SIN signaling and depends on the SIN to be achieved. Thus, we propose that the CIP participates in a checkpoint, capable of triggering a CAR constriction delay through the SIN pathway to ensure that cytokinesis terminates successfully.Amid growing concern about the adverse effects of pesticides, a questionnaire-based cross-sectional study was conducted to evaluate farmers' knowledge on pesticides handling. Results of in-depth surveys (n = 217) with field observations revealed that participants had a mean 18 years of farming experience and knapsack-sprays were used for spraying. About 99% were not using any personal-protective-equipments during mixing, loading, application operations. Over 80% reentered the treated farms within two-days of application without following safety protocols and 39% admitted unsafe disposal of empty containers of pesticides. Statistical-analysis revealed a significant association between participant's pesticide handling practices and self-reported morbidity symptoms (p  less then  0.05). Despite awareness of toxicity, participants were mishandling the pesticides. Training on safe handling practices and incentives in form of personal-protective-equipments could be productive to ensure safety at work.Sustained attention plays an important role in adaptive behaviours in everyday activities. As previous studies have mostly focused on vision, and attentional resources have been thought to be specific to sensory modalities, it is still unclear how mechanisms of attentional fluctuations overlap between visual and auditory modalities. To reduce the effects of sudden stimulus onsets, we developed a new gradual-onset continuous performance task (gradCPT) in the auditory domain and compared dynamic fluctuation of sustained attention in vision and audition. In the auditory gradCPT, participants were instructed to listen to a stream of narrations and judge the gender of each narration. In the visual gradCPT, they were asked to observe a stream of scenery images and indicate whether the scene was a city or mountain. find more Our within-individual comparison revealed that auditory and visual attention are similar in terms of the false alarm rate and dynamic properties including fluctuation frequency. Absolute timescales of the fluctuation in the two modalities were comparable, notwithstanding the difference in stimulus onset asynchrony. The results suggest that fluctuations of visual and auditory attention are underpinned by common principles and support models with a more central, modality-general controller.This study is devoted to investigating mechanisms that inhibit habituated response associated with affordance of a non-target while executing action directed to a target. In four experiments, a paradigm was used that required a rapid left- or right-hand response according to the direction of the target arrow presented simultaneously or in close temporal proximity to a non-target whose handle position afforded grasping with the left or right hand. In general, responding was decelerated and more erroneous when the handle position was compatible with the responding hand. This effect of response inhibition was removed when the delay between the non-target offset and target onset was longer than 200 ms, and reversed into response facilitation when the target onset was delayed for 400-600 ms. The study suggests that processes that control withholding habitual response associated with affordance of a non-target utilise response inhibition mechanisms overlapping with those involved in behavioural control of the stop-signal task. This response inhibition is triggered automatically and directly by affordance of a non-target without preceding response excitation associated with this affordance cue.The Korean writing system has the flexibility of writing horizontally and vertically as well as two syllabic formats that cannot be found in any other alphabetic script. Consolidating these two characteristics, this study investigated the differential extractions of visual information from the mutilated stimuli of the two syllabic formats of CVC syllables and two writing directions using a lexical decision task. In Experiment 1, the two syllabic formats (i.e., balanced syllables, [Formula see text] and vertical syllables, [Formula see text]) were mutilated downwards and upwards (i.e., [Formula see text] [Formula see text] respectively), along with whole stimuli, in horizontal writing direction. In Experiment 2, the same stimuli and syllabic formats were mutilated rightwards and leftwards (i.e., [Formula see text] [Formula see text] respectively) in vertical writing direction. Linear mixed effects models showed significant syllabic format effects and writing direction effects, indicating an upper-part superiority in horizontal writing and a right-part advantage in vertical writing.
