Humans with biallelic inactivating mutations in Epithelial Cell Adhesion Molecule (EpCAM) develop congenital tufting enteropathy (CTE). To gain mechanistic insights regarding EpCAM function in this disorder, we prepared intestinal epithelial cell (IEC) organoids and spheroids. IEC organoids and spheroids were generated from ROSA-CreERT2 EpCAMfl/fl mice. Proliferation, tight junctions, cell polarity and epithelial integrity were assessed in tamoxifen-induced EpCAM-deficient organoids via confocal immunofluorescence microscopy and Western blotting. Olfm4-expressing stem cells were assessed in IEC cells in vitro and in vivo via fluorescence in situ hybridization. To determine if existing drugs could ameliorate effects of EpCAM deficiency in IEC cells, a variety of pharmacologic inhibitors were screened. Deletion of EpCAM resulted in increased apoptosis and attenuated growth of organoids and spheroids. Selected claudins were destabilized and epithelial integrity was severely compromised. Epithelial integrity was improved by treatment with Rho-associated coiled-coil kinase (ROCK) inhibitors without restoration of claudin expression. Correspondingly, enhanced phosphorylation of myosin light chain, a serine/threonine ROCK substrate, was observed in EpCAM-deficient organoids. Strikingly, frequencies of Olfm4-expressing stem cells in EpCAM-deficient IEC cells in vitro and in vivo were decreased. Treatment with ROCK inhibitors increased numbers of stem cells in EpCAM-deficient organoids and spheroids. Thus, EpCAM regulates intestinal epithelial homeostasis via a signaling pathway that includes ROCK.Urinary tract infections (UTIs) are common in clinics and hospitals and are associated with a high economic burden. Enterobacterium Klebsiella pneumoniae is a prevalent agent causing UTIs. A high prevalence of carbapenem-resistant K. pneumoniae (CRKP) has emerged recently and is continuing to increase. Seventeen urinary CRKP isolates collected at a teaching hospital in Taiwan from December 2016 to September 2017 were analyzed to elucidate their drug resistance mechanisms. Two-thirds of the isolates were obtained from outpatients. AZD5363 Antimicrobial susceptibility tests demonstrated multidrug resistance in all the isolates. Multilocus sequence typing analysis showed high diversity among the isolates. PCR analysis demonstrated the presence of carbapenemases in three isolates. All isolates carried at least one other extended-spectrum β-lactamase, including TEM, DHA, and CTX-M. Fifteen isolates contained mutations in one of the outer membrane porins that were assessed. The expression levels of the acrB and/or oqxB efflux pump genes, as determined by qRT-PCR, were upregulated in 11 isolates. Six isolates might have utilized other efflux pumps or antimicrobial resistance mechanisms. This study expands the utilization of two- and three-dimensional detection technologies in underwater applications to detect abandoned tires. A three-dimensional acoustic sensor, the BV5000, is used in this study to collect underwater point cloud data. Some pre-processing steps are proposed to remove noise and the seabed from raw data. Point clouds are then processed to obtain two data types a 2D image and a 3D point cloud. Deep learning methods with different dimensions are used to train the models. In the two-dimensional method, the point cloud is transferred into a bird's eye view image. The Faster R-CNN and YOLOv3 network architectures are used to detect tires. Meanwhile, in the three-dimensional method, the point cloud associated with a tire is cut out from the raw data and is used as training data. The PointNet and PointConv network architectures are then used for tire classification. The results show that both approaches provide good accuracy.Bisphenol A (BPA), a precursor to important plastics, is regarded as a common aquatic micropollutant with endocrine-disrupting activity. In the present study, we explored the capability of a UV KrCl excilamp (222 nm) to degrade BPA by a photo-Fenton-like process using persulfate under flow-through conditions. The first-order rate constants of degradation were obtained and the mineralization of dissolved organic carbon (DOC) was estimated. The results showed complete BPA degradation and high DOC mineralization (70-97%). A comparative analysis of degradation rates and DOC removal in the examined systems (UV, Fe2+/S2O82-, UV/S2O82- and UV/Fe2+/S2O82-) revealed a significant synergistic effect in the photo-Fenton-like system (UV/Fe2+/S2O82-) without the accumulation of toxic intermediates. This indicated that the BPA was oxidized via the conjugated radical chain mechanism, which was based on the photo-induced and catalytic processes involving HO• and SO4-• radicals. The primary intermediates of BPA degradation in the UV/Fe2+/S2O82- system were identified by HPLC/MS and the oxidation pathway was proposed. The high performance of the photo-Fenton-like process employing a quasi-monochromatic UV radiation of a KrCl excilamp offers promising potential for an efficient removal of such micropollutants from aqueous media.Epidemiological studies have reported that consumption of dairy products rich in β-lactolin is beneficial for cognitive decline among elderly individuals. Although previous studies have shown that β-lactolin supplementation improves memory function and attention in healthy adults, the mechanism through which β-lactolin affects human brain function has yet to be elucidated. This placebo-controlled randomized double-blind study therefore examined the effects of β-lactolin on human regional cerebral blood flow (rCBF) using near-infrared spectroscopy (NIRS) according to the Consolidated Standards of Reporting Trials guidelines. A total of 114 healthy participants aged between 50 and 75 years with relatively low cognition were randomly allocated into the β-lactolin or placebo groups (n = 57 for both groups) and received supplementation for 6 weeks.
