•The parameters of the method are adjusted using both observers opinion and the well-known reference image quality assessment BRISQUE (Blind/Referenceless images spatial quality Evaluator) score. © 2020 The Authors.Hydrogen peroxide (H2O2) is an important signal molecule produced in animal and plant cells. The balance of H2O2 between the intra- and extracellular space is regulated by integral membrane proteins, which thereby modulate signaling. Several methods have been established to analyze aquaporin mediated transport of H2O2 in whole cells with the intrinsic limitation that the amount of protein responsible for a certain activity cannot be standardized. As a consequence, the quantification of the transport and specific activity is difficult to extract making it problematic to compare isoforms and mutated variants of one specific target. Moreover, in cell-based assays, the expression of the target protein may alter the physiological processes of the host cell providing a complication and the risk of misleading results. To improve the measurements of protein based H2O2 transport, we have developed an assay allowing quantitative measurements.•Using purified aquaporin reconstituted in proteoliposomes, transport of H2O2 can be accurately measured.•Inside the liposomes, H2O2 catalyzes the reaction between Amplex Red and horseradish peroxidase (HRP) giving rise to the fluorescent product resorufin.•Analysing pure protein provides direct biochemical evidence of a specific transport excluding putative cellular background. © 2020 The Author(s).Palygorskite is a kind of crystalline hydrated magnesium aluminum silicate mineral with micro-fibrous morphology. Due to the large specific surface area, moderate cationic exchange capacity and pronounced adsorption properties, it has been widely used in many fields. In order to enhance the loading capacity and adjust the microstructure of palygorskite (Pal) crystal, series of three-dimensional palygorskite carriers (3D Pal) with different pore size were fabricated through grafting from or grafting onto method. Due to the functional and cross-linked molecules act as upholder reagents, the specific surface of individual palygorskite is fully utilized and the load capacity is greatly improved. The porosity and pore size of 3D palygorskite based carrier also can be regulated by the length of organic molecular chain segments. The successful preparation of 3D Pal-based carrier provides a new way for surface grafting, modification or preparing 3D carrier of palygorskite and other minerals.•The developed method allows fabricating three-dimensional palygorskite based carriers by covalent bonding method.•The pore size of the as-prepared carriers can be conveniently adjusted by length of the bonded molecular chain. © 2020 The Authors.A quantification assay based on isotope dilution mass spectrometry to determine the concentration of progesterone in human serum was reported. Incorporated with 13C3-progesterone, serum samples were subjected to progesterone extraction and clean-up by C4 solid-phase-extraction columns and hexane-based liquid/liquid extraction, respectively. The cleaned-up serum samples were then subjected to MALDI-TOF mass spectrometry for the quantification of progesterone. In the study, the recovered progesterone concentration determined by the assay showed good robustness and constancy in comparison to conventional radioimmunologic assay. We concluded that the 13C3-progesterone-based quantification assay is a robust method for the measurement of serum progesterone. Advantages of this technique includes • This study describes a MALDI-TOF/MS method for the determination of serum progesterone. • The technique is simple and easy to apply on MALDI-TOF/MS for serum progesterone analysis. • The correlation coefficient between MALDI-TOF MS and RIA was 0.981 for serum progesterone. learn more © 2020 The Authors. Published by Elsevier B.V.A new risk assessment method intended for comparing long-term environmental performance of different types of sanitary landfills was customized. Processes occurring within the hydrogeological environment were excluded from modeling, because they can be addressed separately. Only parameters directly related to leachate composition at the bottom of the landfill and leachate losses into the subsoil after landfill closure which can be reliably determined by evaluating already available information from the scientific literature were considered as necessary inputs for quantitative modeling. Once the simulated outcomes for a primary output ''fugitive emissions of a reference pollutant into the subsoil'' are acquired, more complex outputs can be derived, too. Commercially available risk assessment software which operates within an Excel environment was used to fulfill the task.•Uncertainty of data as well as heterogeneity and complexity of landfill systems was considered by attributing the selected input parameters with adequate probability density functions•Probability density functions attributed to the inputs differ considerably between the antagonistic landfill types•Risk assessments related outputs were defined as probabilities that an aquifer would be polluted due to landfill derived emissions into the subsoil. © 2020 The Author(s).Domestication and cultivation of tree species, such as Pongamia pinnata is quite important because of its biofuel properties. Seedlings grown in modified hydroponic culture were morphologically similar to that of soil grown seedlings. Further, seedlings were allowed to grow without root limitation. Comparatively, our modified hydroponic growth system can be performed with minimal resources. Prior incubated root segments with CoroNa-Green AM dye retained maximum amount of dye when compared to CoroNa-Green AM dye incubated sections. Our modified protocol provides quantitative analysis of 2D and 3D imaging process at cellular and sub-cellular level.•Our protocol is customized to study individual plant behavior.•Additionally, it is customized for growing tap rooted trees species hydroponically. Changing the nutrient solution with regular intervals provides continuous supply of nutrients to the plants.•Prior incubation of root segments with Na+ probe (CoroNa-Green AM) provides better resolution in imaging process.
